The relationship between expression of CD147 and PHH3 in invasive breast cancer and clinical pathological features

Abstract
Objective To observe the quantitative expression of CD147 protein and the number of PHH3 positive cells in invasive breast cancer,and analyze their relationship and correlation with multiple clinicopathological characteristics. Methods 80 cases of primary invasive breast cancer surgical specimens were selected,and 29 cases of normal breast tissues were selected as the control group. The expression of CD147 and PHH3 was detected by immunohistochemical SP method. The expression of CD147 protein was quantitatively detected by IPP6.0 image analysis software,and the number of PHH3 positive cells and mitotic index was counted. The relationship between the expression of CD147 and PHH3 in invasive breast cancer and its clinicopathological characteristics was analyzed. Spearman method was used to analyze the correlation between the two. Results CD147 was mainly located in the cell membrane or membrane plasma of breast cancer cells,and PHH3 was mainly located in the mitotic nuclei of invasive breast cancer. The quantitative expression and positive number of both in invasive breast cancer tissue were significantly higher than those in normal breast tissue(P< 0.000). In 80 cases of invasive breast cancer,the quantitative expression of CD147 protein was higher in larger tumor diameter,higher WHO grade,lymph node metastasis,TNM Ⅲ-Ⅳ stage,estrogen receptor (ER) negative,Her-2 gene without amplification (P< 0.05);The number of PHH3 and mitotic index was higher in larger tumor diameter,higher WHO grade,lymph node metastasis,and TNM staging stages III-IV (all P< 0.05). The higher the expression of CD147 protein,the more PHH3 cells. Spearman analysis showed a significant positive correlation between CD147 and the number of PHH3 positive cells and mitotic index (r=0.950,r=0.706,P=0.000). Conclusion The high expression of CD147 and PHH3 in invasive breast cancer is related to the occurrence and development of tumors. They are positively correlated. Detecting the quantitative expression of CD147 protein,the number of PHH3-positive cells,and mitotic index in breast cancer helps to the judge invasion and metastasis,and provide reference value for clinical and pathological diagnosis of breast cancer.
